Real-Time Inhalation Analytics Using Single Particle Aerosol Mass Spectrometry (SPAMS)

Abstract:

Single particle aerosol mass spectrometry (SPAMS) enables the real-time determination of aerodynamic particle size and chemical composition of inhaled pharmaceuticals. The aerodynamic diameter and a dual-polarity laser time-of-flight mass spectrum of up to 250 individual particles are measured per second. SPAMS analyses performed on a commercial formulation of indacaterol demonstrated analytical consistency in repeated measurements carried out over a two week period. The particle size distribution of aerosolized ciprofloxacin determined using SPAMS analysis was compared with cascade impaction data to enable scaling between this novel technique and the standard regulatory and pharmacopeial approach. The SPAMS technique was shown to be reproducible and the data found to be related to the results of Next Generation Impactor (NGI) analyses coupled with high performance liquid chromatography (HPLC).
