A Physiologically-based Pharmacokinetic Model to Predict Nose-to-Brain Drug Delivery

Abstract:

This study investigated the delivery of sumatriptan to the brain via the nose using a combination of in vivo and in silico methods. A non-clinical non-human primate study and a physiologically-based pharmacokinetic (PBPK) model revealed that nasal formulations significantly increased drug transport to the brain due to enhancement of permeation rather than greater deposition in the olfactory region. A sensitivity analysis of the PBPK model highlighted the importance of optimizing nasal epithelial permeability and direct nose-to-brain transport rates. The findings highlight the potential of mechanistic models to enhance our understanding of nose-to-brain drug delivery and improve the development of products for central nervous system (CNS) diseases.
