21st Century Particle Engineering: Particle Design, Manufacture and Scale-up of PRINT® Technology

Abstract:

Particle replication in non-wetting templates (PRINT®) technology (Liquidia Technologies, Research Triangle Park, NC, USA) is a unique particle engineering approach that enables the design and scalable manufacturing of particles for pharmaceutical products. Because of the precise control of size and shape and compatibility with small and large molecules, PRINT technology is well suited to address the formulation and delivery requirements for inhalation products. PRINT technology utilizes advanced lithographic techniques to design a mold template. PRINT particles are formed by filling the cavities of the mold template with API or formulated drug product. “Roll-to-roll” manufacturing, involving coating, molding, and harvesting, is used to produce PRINT particles at sufficient scale for pharmaceutical products. Multi-kilogram scale has been achieved to support the non-clinical, clinical, and planned commercial activities. This article reviews the design, manufacture and scale-up, as well as emerging application areas for PRINT particles.
